Procedure and Interpretation of the #Rivalta's Test
Rivalta's Test Standard Procedure
• A plastic test tube (volume 10 mL) is filled with 7 mL of distilled water
• 20 mcL (1 drop) of acetic acid (100%) is added
• The solution is thoroughly mixed
• 20 mcL (1 drop) of the effusion is layered on the surface of this solution
• The outcome of the drop is observed
Rivalta's Test Interpretation
• The Rivalta's test is defined as positive
• If the drop stays attached to the surface
• If the drop retains its shape and keeps a connection to the surface
• If the drop slowly floats down to the bottom
• If the drop does not stay in shape, but changes to an “upside-down jelly fish”
• If the drop disperses in little cloudy pieces that can be observed falling down to the bottom of the tube
• The Rivalta's test is defined as negative
• If the drop disappears and the solution remains clear
• If clear or cloudy cords seen are falling down, which dissolve before reaching the bottom of the tube,
but no drop or “upside-down jelly fish” is visible

1. Microbial Fermentation: Ruminants have a specialized compartment in their stomach called the rumen, which houses billions of microorganisms, including bacteria, protozoa, and fungi. These microorganisms have the ability to break down complex carbohydrates, such as cellulose and hemicellulose, present in plant materials.
2. Amino Acid Synthesis: During microbial fermentation in the rumen, the microorganisms produce enzymes that break down plant proteins into simpler forms, including amino acids. Ruminants can then absorb these amino acids synthesized by the rumen microbes and utilize them for their own protein synthesis.
3. Recycling of Nitrogen: Ruminants have a unique ability to recycle nitrogen. When proteins are broken down in the rumen, the resulting ammonia is converted by the rumen microbes into microbial proteins. These microbial proteins are then digested and absorbed in the lower parts of the digestive system, providing the ruminant with essential amino acids.
4. Protein Synthesis in the Rumen: The rumen microbes themselves are a rich source of protein. When ruminants digest the microbes in the lower parts of their digestive system, they obtain essential amino acids from the microbial proteins.
Due to these mechanisms, ruminants can meet their essential amino acid requirements through microbial fermentation and recycling of nitrogen, without relying on dietary sources of essential amino acids. However, it's important to note that ruminants still require a balanced diet to meet their energy, vitamin, and mineral requirements.
